Quantitation of Western blots from polyacrylamide gels is commonly accomplished by either visual inspection or densitometry. However, due to absorption of protein and staining reagents by the blotting membrane it is possible that visual inspection and reflectance densitometry will given anomalous results. In this paper we show such an example, and discuss how transmission densitometry can provide more accurate results.
