Type I IFNs are central to a vast array of immunological functions. IFN-alpha/beta production is initiated at the early stages of the innate immune response and, therefore, it is likely to be the dominant factor in shaping downstream events in the innate and adaptive immune responses. Indeed, the major role of IFN-alpha is the induction of the priming state through the production and regulation of other mediators, including cytokines (i.e., IL-6 and IL-15), the up-regulation of MHC and costimulatory molecules, chemokines and chemokine receptors. The main objective of the present study was to determine the involvement of IFN-alpha on murine CD4+CD25- T helper (Th) cell activation, as well as to define the role played by this cytokine on CD4+ CD25+ regulatory T (Treg) cell proliferation and function. Although IFN-alpha promotes CD4+CD25- Th cells co-incubated with APCs to produce large amounts of IL-2, the ability of these cells to respond to IL-2 proliferative effects is prevented. Moreover, in medium supplemented with IFNalpha, IL-2 induced CD4+CD25+ Treg cell proliferation is inhibited. Notably, IFN-alpha also leads to a decrease of the CD4+CD25+ Treg cell suppressive activity. Altogether, these findings indicate a dual pivotal role of IFN-alpha¡ on CD4+CD25- Th cell activity by a direct control on APCs activation and indirectly by affecting CD4+CD25+ Treg cell-mediated suppression. Moreover, we also analysed if IFN-alpha could affect in vitro Treg cell differentiation after polyclonal stimulation of naive T cells in the presence of TGF-beta and IL-2. IFN-alpha, synergizing with TGF-beta and IL-2, might influence the Treg/Th-17 balance, leading to almost total conversion of naive T cells in CD4+CD25+Foxp3+ iTreg, sustaining both Foxp3 and CD25 expression and reciprocally inhibiting Th-17 cell differentiation.

Gli IFN di tipo I sono coinvolti in diverse funzioni immunologiche. La produzione di IFN-alfa/beta è indotta nelle prime fasi dell' immunità innata e successivamente diviene un fattore dominante nella risposta innata e adattativa. Infatti l'IFN-alfa/beta ha un ruolo fondamentale nell'induzione di uno stato di attivazione, attraverso la produzione e regolazione di mediatori, come le citochine (ad esempio IL-6 e IL-15), l' aumento dell' espressione dell' MHC e delle molecole costimolatorie, chemochine e recettori per chemochine. L'obiettivo principale del presente studio è stato determinare il ruolo dell'IFN-alfa nell' attivazione delle cellule CD4+CD25- T helper (Th) murine e l' effetto di questa citochina sulla proliferazione e funzionalità delle cellule CD4+ CD25+ T regolatorie (Treg). Sebbene l' IFN-alfa promuova nelle cellule Th CD4+CD25- co-incubate con le APC la produzione di elevati livelli di IL-2, tuttavia queste cellule non proliferano in maniera comparabile alla quantità di IL-2 presente nel mezzo. Inoltre in presenza di IFN-alfa, la proliferazione delle cellule Treg indotta da IL-2 risulta inibita. In particolare l'IFN-alfa inibisce l' attività soppressoria delle cellule Treg CD4+CD25+. I risultati suggeriscono quindi che l' IFN-alfa esercita un duplice ruolo sull' attività delle cellule Th CD4+CD25-, attraverso un controllo diretto dell 'attivazione delle APC e indirettamente influenzando la soppressione mediata dalle cellule Treg CD4+CD25+. E' stato inoltre analizzato se l'IFN-alfa possa condizionare il differenziamento in vitro delle cellule Treg, in seguito a stimolazione policlonale delle cellule T naive in presenza di TGF-beta e IL-2. I risultati dimostrano che l' IFN-alfa, agendo in sinergia con il TGF-beta e IL-2, influenza il balance Treg/Th-17, inducendo una quasi completa conversione delle cellule T naive in cellule iTreg CD4+CD25+Foxp3+, sostenendo l' espressione sia del CD25 che di Foxp3 e inibendo reciprocamente il differenziamento delle cellule Th17.
